Hi,
I am having a problem with Mellel. When I change a format of a paragraph using the paragraph style I lose all the character variation formats (bold, italics...) is there a way to avoid this?

Are you using style variations, or are you making the formatting changes with Cmd+[B,I,&c.]? If you're not using style variations, then this is your problem.

Hum... I have a problem. I am importing RTF files from Scrivener. Is there a way to find all Italics in text and replace them with the format using character variation?

Hi,
you could replace styled words with a character style and/or character style variation by using the File > Replace Syles… comand. It’s somewhat hidden but works very well. There is also a way to do the same via the powerful Find&Replace feature but I think the the Replace Styles… thing is easier to use.
